  • Sujet: Polysorbates versus hydroxypropyl-betacyclodextrin (HPBCD) stability comparative study and their monoclonal antibody stability benefits
    Polysorbates are intensively used in biologics formulation as stabilizer due to their propensity to inhibit aggregation at interfaces but possesses low intrinsic stability. This study is focusing on bringing to formulator attention, as an alternative, a new class of molecules hydroxypropyl-betacyclodextrin (HPBCD). When subjected to various physical and chemical stressors: heat, light and oxidative stresses, HPBCD maintains its chemical stability with >97% recovery while polysorbate showed significant degradation. For adalimumab, as a model mAb, formulated in presence of HPBCD we observed a significant decrease in both agitation and thermal stress-induced protein aggregation and sub-visible particle formation prevention compared to polysorbate.

Dr. Hailong Zhang is analytical research manager in applied science at Roquette. Dr. Zhang received his Ph.D. in Analytical-bioanalytical Chemistry at Cleveland State university /Cleveland Clinic and completed postdoctoral training in pharmaceutical science at University of Toronto. Dr. Zhang’s interests and experience focus on exploring phase-appropriate analytical strategies to the specific needs of unique molecules, delivering a comprehensive understanding and characterization of the molecule for each stage of drug development and commercialization, including starting material, intermediates and drug products under cGMP.
